The SPRIworks Fragment Library System I method begins with the off-line shearing of input DNA by nebulization or other means. Fragmented DNA is then loaded onto the SPRI-TE instrument which performs the subsequent steps in the library construction process.
The SPRIworks Fragment Library System I employs enzymatic reactions, SPRI-based reaction purifications, and
optional SPRI-based size selection to automatically generate fragment libraries. All reagents required to construct a single library are contained within the reagent cartridge. Additional reagents and primers are required for subsequent enrichment of the library. The method can generate from one to ten libraries simultaneously per 5-hour run.
After processing on the SPRI-TE instrument, libraries are further amplified and used as templates for sequencing on the Illumina Genome Analyzer.
